Composition

Detergent solution in distilled water coloured with phenol red.

Test principle

The reagent gives a positive reaction if mixed with milk containing multiplied cell elements (nuclear in particular) the actual reaction of which (pH value) is changed.
The milk shows the above mentioned changes particularly after the irritation of the mammary gland parenchyma due to both bacterial and nonbacterial influences.
Changes in consistency and the forming of a gel of varying degrees of viscosity can be observed when the preparation is mixed with milk with an increased volume of nuclear cell elements.
The colour of the mixture changes simultaneously according to the change of the pH value of the milk.
A yellow colour means an acidic reaction and a red colour means a basic reaction.

Indication

Diagnostic for dairy cows suspected having mastitis, as a quick stable test.

Target animals

Cattle.

Testing

Mix approx. 2 ml of milk fresh from the cow with 2 ml of the test solution on dishes of the evaluating pallet; evaluate the reaction whilst slightly tilting the pallet; the onset of the reaction usually begins within 30 seconds. Evaluating pallets can be supplied by Bioveta by request. Pallets are made of plastic and each dish has an adjustment line of 2 ml.

Procedure for milk admeasurement on a dish
  1. Draw some milk from each udder to respective pallet dish.

  2. Adjust the volume of the milk to 2 ml by tilting the pallet to pour off some milk.

Mastitis test NK dosing procedure by means of a pump
  1. Unlock the pump from transport position to position OPEN

  2. Spray the first dose after unlocking outside the pallet (the pump is being filled)

  3. Every consecutive single squeeze = 2 ml of the product

  4. Dose 2 ml of the product (one squeeze of the pump) to each dish

Test evaluation

With a positive reaction, the characteristic formation of flocks, fibres or a gel of various intensity can be observed. With a strong positive reaction, the gel gathers in the centre of the dish after the mixture sets. A colour reaction appears simultaneously due to the effect of the different pH values on the colour indicator. A positive reaction can be observed if the amount of cell elements exceeds 200,000 ml/1 ml of the mixture.

Note

A positive reaction can also be observed in milk from healthy cows, namely, at the onset of a lactation period (up to 3 - 5 days, sometimes even longer) and shortly before delivery. The same reaction can also be observed also during the rutting season or due to a sudden change of feed. Reactions from all 4 quarters usually show the same intensity in the above-mentioned cases.

Storage

Store at temperatures not exceeding 25 °C. The product must not freeze! The product must not be used after the expiry date which is stated on the packaging.
